Africa Special with Karen Hogg (University of York) and Margaret Mbuchi (KEMRI)

Flow Stars host Peter O'Toole discusses Flow Cytometry in Africa with  Karen Hogg at the University of York, and Margaret Mbuchi at the Kenya Medical Research Institute. If you want to get involved and support research in resource-poor areas, find out how to donate spare instruments via the International Society for the Advancement of Cytometry Instruments 4 Science program.

Combining CRISPR with Flow-FISH to Study CRISPR-Mediated Genome Perturbation

Since the advent of the clustered regularly interspaced short palindromic repeats (CRISPR)–CRISPR-associated (Cas) system as a genome editing tool, the ease of studying gene function and the impact thereof on cellular function has increased incrementally. Not surprisingly, the original describers of the CRISPR/Cas system received the 2020 Nobel Prize in Chemistry.

Rapid Purification and Multiparametric Characterization of Circulating Small Extracellular Vesicles Utilizing a Label-Free Lab-On-A-Chip Device

This paper describes a new insulator-based dielectrophoretic device to rapidly isolate small extracellular vesicles from biofluids and cell culture media, based on their dielectric properties. Yield and purity of sEVs isolated using the iDEP device were found to be comparable to techniques including DU and SEC, the platform overcomes some of the key limitations in EV isolation by having added advantages.

Explainable Machine Learning for Profiling the Immunological Synapse and Functional Characterization of Therapeutic Antibodies

Therapeutic antibodies are widely used to treat severe diseases. Most of them alter immune cells and act within the immunological synapse; an essential cell-to-cell interaction to direct the humoral immune response. Although many antibody designs are generated and evaluated, a high-throughput tool for systematic antibody characterization and prediction of function is lacking.

Exploring The Single Cell Immune Landscape Of Renal Allograft Inflammation Using Imaging Mass Cytometry

pubmed.ncbi.nlm.nih.gov

Kidney allograft inflammation, mostly attributed to rejection and infection, is an important cause of graft injury and loss. Standard histopathological assessment of allograft inflammation provides limited insights into biological processes and immune landscape. Here, using Imaging Mass Cytometry...

High-Dimensional Multi-Pass Flow Cytometry via Spectrally Encoded Cellular Barcoding

www.nature.com

Advances in immunology, immuno-oncology, drug discovery and vaccine development demand improvements in the capabilities of flow cytometry to allow it to measure more protein markers per cell at multiple timepoints. However, the size of panels of fluorophore markers is limited by overlaps in fluorescence-emission spectra, and flow

MetaGate: Interactive Analysis of High-Dimensional Cytometry Data with Meta Data Integration

pubmed.ncbi.nlm.nih.gov

Traditionally, data analysis is carried out using manual gating, in which cut-offs are defined manually for each marker.  To tackle the  escalation in data complexity, numerous new analysis algorithms have been developed.. We developed MetaGate as a platform for...

Interpretable Unsupervised Learning Enables Accurate Clustering With High-Throughput Imaging Flow Cytometry

www.nature.com

A primary challenge of high-throughput imaging flow cytometry (IFC) is to analyze the vast amount of imaging data, especially in applications where ground truth labels are unavailable or hard to obtain. We present an unsupervised deep embedding algorithm...
